This trip around New York is best done in two days to get the most out of the city. If you don't find anything to do then you're not looking hard enough. Roam the streets but PLEASE stay away from Time Square the best of the city exists far far away from it.
For your trip I would recommend you focus on Soho, China Town, Queens, Tribeca, East Village, and if you get a chance to go over the Brooklyn bridge - DO.
I usually start my trips in Little Italy where I grab some wonderful lunch and then roam around the endless concrete jungle that is New York. The best way to experience the city is choose the area that sounds most interesting to you and walk until you get lost. Something as simple as walking around could result in an amazing day because there's something wonderful waiting for you on every corner; restaurants, bars, poetry clubs, comedy clubs - you name it its here.
When it comes to transportation the train is the cheapest transportation in the city when you're traveling a somewhat long distance. At the end of the night when you're tired and just want to get to your hotel I suggest the cab no matter how much cheaper the train would be, It's less of a hassle and you get to see more of the city out your window.
Before you catch that cab finish the night with some great food and tasty drinks at a hidden bar deep within Brooklyn or East Village. Oh, at this point take a cab home no matter how much cheaper the train would be, It's less of a hassle and you get to see more of the city out your window.
